Il fut le fils d'Annibale Ier et de Donnina, la fille de Lancillotto Visconti.
1443, il naquit à Bologne.
1464, il épousa la veuve Ginevra Bentivoglio, la fille illégitime d'Alessandro Sforza.
Il fut condottière offrant ses services au duché de Milan, à la république de Florence.
Sous son impulsion, Bologne prit un nouvel essor politique, artistique et civil, mais il fut le dernier prince de Bologne.
1506, le pape Jules II désirant annexer Bologne aux territoires de l’Église l'excommunia et le chassa ; la noble famille des Bentivoglio fut précipitée dans la ruine.
1508, il mourut prisonnier du roi de France Louis XII dans un cachot du château des Sforza du duché de Milan.
1508, il est enterré à Bologne dans l'église San Giacomo Maggiore ou église Saint-Jacques-le-Majeur.
